Jena Alliance course: Preparation of Peptide Samples for LC-ESI-MS/MS Analysis
Theme of the workshop:
The workshop deals with the preparation of peptide samples for liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). We will provide SDS-polyacrylamide gels loaded with protein crude extracts from Chlamydomonas reinhardtii and show the techniques which lead to a successful identification of peptides from gel bands using high-resolution mass spectrometry. Furthermore, the theoretical background of each method will be explained.

- Preparation of SDS-polyacrylamide gels suited for mass spectrometry analyses
- Excision of gel bands and destaining procedures
- In-gel tryptic digestion and elution of peptides
- Reversed phase chromatography (using ZipTips®)

The number of participants will be limited to 6 in order to assure a workshop environment conducive to “hand to hand” exercises. Students only need a basic background in SDS-PAGE procedures, principles of reversed phase chromatography and mass spectrometry.
